Understanding HPLC Purity Documentation
How high-performance liquid chromatography (HPLC) purity results are generated and documented, what an area-percent value does and does not represent, and which method parameters give a purity figure its meaning.
What HPLC measures
High-performance liquid chromatography separates the components of a sample as they travel through a column at different rates. A detector records each component as it elutes, producing a chromatogram of peaks over time. Purity by HPLC is typically reported as the area of the target peak expressed as a percentage of the total integrated peak area.
This "area-percent" figure describes the relative response of the target compound under the specific detection conditions used. It is a documented, reproducible measurement — but its interpretation depends on the method that produced it.
Why the method parameters matter
An area-percent value is only interpretable alongside the conditions that generated it. A complete purity record identifies the detection and separation parameters so the figure can be understood and, in principle, reproduced.
- Detection wavelength — UV detection at a stated wavelength (for example 214 nm) responds differently to different chromophores.
- Column and mobile phase — the stationary phase and gradient determine what is separated from what.
- Run time and gradient — determine whether late-eluting components are captured.
- Integration parameters — define how peak areas are assigned.
What an area-percent value does not tell you
Area-percent by a single detector is a relative measure of detector response, not an absolute mass fraction. Components that do not absorb at the detection wavelength contribute little or no signal, so they are not represented in the percentage. For this reason, purity by HPLC-UV is one line of evidence that is strongest when read together with an orthogonal identity method such as mass spectrometry and, where relevant, water-content or residual-solvent determinations.
Reading purity documentation well means reading the whole method context, not a single number in isolation.
How purity appears on a Certificate of Analysis
On a COA, an HPLC purity result is generally stated as an area-percent against a named method, alongside the report date and the reviewer who approved the document. Where a supplier provides the underlying chromatogram, the reported value can be traced back to the integrated peaks that produced it.
A purity figure without its method context should be treated as incomplete rather than as a finished result.
